Thank you tjohnson_nb. Excellent overview. Some points I am mulling over.

The connection between resveratrol and calorie-restriction was interesting!

I thought they may have confused the term "aging" with "life span" during the discussion of the antioxidant experiments.

The idea that the childhood aging disease progeria was not telomere shortening - but an abnormal protein was also interesting. Wonder if that has been confirmed.

The centenarians was an interesting segment, and in addition to their 3 genes, one wonders if their intracellular glutathione levels were higher - as per Levy's GSH MASTER DEFENDER revelations.

Bruce Ames refers to Tomas Prolla's work. This video on calorie restriction - trying to understand it - is fascinating. Near the end they report the finding that intracellular glutathione (GSH) universally (all tissues) at least DOUBLES under calorie restriction.

Summary so far.

#1. For immortality (at least life-span increase) telemore shortening has to be dealt with. If telomerase preferentially lengthens the shortest telomeres, then it is key. (Human blood testing for report on the shortest telomers: http://lifelength.com If anyone finds cost/lab please post.)

#2 For Anti-Aging/Age reversal, calorie restriction is such a profound anti-aging mechanism, that it prevents age-related hearing loss - which can be experimentally measured in lab mice. No one wants to restrict calories so the search is on for what genes are expressed during calorie restriction. A laboratory set up to measure gene expression to measure whether drugs/supplements have an effect that mimics calorie restriction http://www.lifegentech.com

The team who set up this lab have duplicated Bruce Ames findings of Acetyl-L-Carnitine and Lipoic Acid (but also found Lycopene, CoQ10 and resveratrol also cause gene expression mimic'ing calorie restriction.)

#3 Vitamin C and perhaps especially Glutathione (GSH) is (are) intimately involved.

Dr. Levy found GSH research that a common denominator of people who live to be over 100 is high levels of glutathione in their cells. Yesterday, in the youtube video the Wisconsin team found that calorie restriction AT LEAST DOUBLES  glutathione levels in all tissues.

According to professor Emeritus of Chemistry Boyd Halley, high dose vitamin C is the only "proven" way to increase intracellular glutathione (GSH).

In the Bruce Ames ALC/ALA studies of these substances that can reverse aging in mice - a finding was increased levels of vitamin C in tissues.

We have at least two studies showing vitamin C reduces the shortening of telomeres, and one study in stem cells! Which reminds me.

The mice subjected to the gene therapy now under test are likewise free of cancer. ...

This study is viewed primarily as "a proof-­‐of-­‐principle that telomerase gene therapy is a feasible and generally safe approach to improve healthspan and treat disorders associated with short telomeres," state Virginia Boccardi (Second University of Naples) and Utz Herbig (New Jersey Medical School-­‐University Hospital Cancer Centre) in a commentary published in the same journal.

We have bitten the bullet :D We just placed the first order for what may very well be the world's first liposomal telomerase activator. The formula also contains molecules that have been shown to mimic calorie restriction's effect on genes and reverse aging in mice. In short, the unique liposomal formula is what I wanted for myself.

The bet is liposomal will work better than pills. The success of our first (China-FREE Quali-C) liposomal vitamin C has exceeded our wildest expectations. I am speaking of success against infections, i.e., the product works, and this has resulted in high sales volume. And as reported at this forum, the liposomal product has been picked up by Vitamin Shoppe as a new product for 2015.

Our experience with truly liposomal vitamin C has me more convinced that ever of Levy's contention, or empirical observation really, that clinically a truly encapsulated vitamin C exceeds the effectiveness of intravenous vitamin C directly into the vein. We are not sure why, but we know it isn't merely a matter of blood concentrations. It is our hope that the dosage amplification effect of delivering nutrients to the cytoplasm (and or nucleus) will allow the small (and thus less expensive dosages) to have a larger anti-aging and telomere lengthening effect than the pill form.

The first batch of the product will be labeled as an Inteligent*Vitamin*C product, but we may form a new company to produce and market follow-on batches. We are trying to find a way for early users to have their blood drawn and telomere length measured before and while taking the product. Ideally, Life Extension would offer the correct blood tests. We have communicated with LEF , but so far they seem uninterested. Another option is to try to work with the telomere measurement lab we have in mind, but we unsure how to obtain the “doctors order” script for the blood testing.

Since this first batch is a sort of Beta test, we will not be charging the full price, until we have established the efficacy of the product, so long as the consumer agrees to the blood tests. Therefore, early users must agree to blood testing for the discount, and perhaps sign an agreement with us. We expect that they will be able to purchase the product at least half the expected retail price of a proven product. We may have to adjust the formula in the future based on these initial measurements. (Note: We are hoping (and working) to be able to deliver the actual human telomerase enzyme itself in the future.)

If we are close to hitting a home run, imagine what kind of world we'd live in if Albert Eistein, Linus Pauling, Isaac Asimov and other geniuses were living and still contributing their combined knowledge and intelligence? Not to mention your favorite athletes extending their playing days into their fifth or six decade, etc

Thank you johnwen - we'll take a look. Bill Andrews recommends lifelength.com - out of Spain - as they measure the shortest telomeres (under the theory we don't care as much about the longer ones?) We are in the process of contacting them. (Ouch thanks for those links. That is the first time we saw the price for lifelength - but if it is the only one to consider, the so be it.)

We are making a huge assumption about what is inert and what isn't, in TA-65. Logically, given the expense of all three molecules lab tests have uncovered (that we now know about thanks to you!) It appears what their saying is that it don't slow aging but enhances Healthy Aging. Which isn't all that bad either!
I included Bill Andrews video (part 1 there is a link to Pt.2) of him talking about it. I didn't watch it probably tomorrow?? The other link gives an assessment of Ta-65 and the last one is the work Hf Valenzuela whose one of the author's of the previous article and his other works.

Terrific links, again thank you johnwen. What I think by what LEF initially told me, is that the telomere tests are so specialized that they required a doctor with knowledge of the results? In a way admitting that their staff can't handle the telomere measurement results. And perhaps there is some fear, maybe with the new Federal laws, of offering these tests.

They do call themselves "Life Extension" and could hire a physician who can evaluate the results, we would hope.

With this in mind, and the idea of "crowds" perhaps having an effect, if readers of this topic were to take action, they may begin to see the value in offering such blood tests!

So good readers, why not contact Life Extension and ask why they aren't offering telomere length lab measurments - even saliva telomere testing?

Yep! I noticed this about LEF they used to be a good source but last couple of years it became apparent their moving towards the light of Pharma’s Money$$!!

So since it’s obvious by this posting your studying senescence (ageing & cell ageing) I figured maybe I should give you a little closer look at some of the other aspects. Things like the Hayflick Limit or premature senescence, replicative senescence all need to be considered as while as those diploid fibroblasts.

As Spock might say, Fascinating!
